Mutation detailsThe endogenous locus was targeted with two rounds of homologous recombination that inserted a floxed neomycin selection cassette into intron 1 and a cassette containing a floxed puromycin resistance gene followed by the lacZ reporter gene fused to a nuclear localization signal (nls-lacZ) into the 3' untranslated region. Exons 2 through 9 were flanked by the floxed selection genes. Cre meditated recombination removed both selection cassettes and exons 2-9. Western blot analysis confirmed the absence of protein in brain lysates of P20 homozygous mutant mice. (J:210236)
