Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 301 (MRL/MpJ x CAST/EiJ)F2 intercross animals to identify QTLs associated with wound healing. Ear hole punch was inflicted on 6 week old animals and wound healing was assessed via ear hole closure. Parental strain MRL/MpJ can fully heal ear hole wounds within 30 days whereas parental strain CAST/Ei cannot. In general female animals are better healers than males (50% faster healing.) Genome scan was performed at an average resolution of 9.4 cM.

Previously identified QTL Heal2 (male specific locus) at 10 cM on mouse Chromosome 13 was confirmed in this study at D13Mit16. Heal2 is a broad peak and may contain several separate QTL peaks. One of these peaks reached significant linkage at 35 cM and is designated Heal7 in the previous study.

03.04.2015 Curators Note: Because Heal2 and Heal7 were originally mapped using an (MRL/MpJ-Faslpr x C57BL/6NTac)F2 intercross in J:50111 in 1998, which differs from the cross used here, we consider this a separate map study and have named these QTL Heal23 and Heal24 respectively.

Heal23 mapped to mouse Chromosome 13 with peak marker D13Mit16 (10 cM), LRT=9.7 in analysis of both male and female mice. Heal23 was more significant to male healing than to female healing and displays an additive effect with homozygous MRL/MpJ mice as the best healers.

Heal24 mapped to mouse Chromosome 13 with peak marker D13Mit13 (35 cM), LRT=18.0. This QTL also displayed sexual dimorphism with more significance in male than in female mice.

A suggestive female specific locus mapped to 55 cM on mouse Chromosome 9 at D9Mit182 (LRT=10.6). MRL/MpJ-derived alleles confer overdominant inheritance at this locus with heterozygous female animals exhibiting the best wound healing. Confirmation of this locus is required. Smad3 is suggested as a possible candidate gene. This locus may overlap with previously identified QTL Stheal9 at 43 cM.

Linkage of male and female wound healing map to slightly different peaks for Heal10 (55 cM) previously identified on mouse Chromosome 11. Peak linkage to female wound healing maps to 60 cM and peak linkage to male wound healing maps to 70 cM. Further analysis is required to determine if Heal10 is indeed two separate male- and female-specific QTLs.

03.04.2015 Curators Note: Because Heal10 was originally mapped using an (MRL/MpJ-Faslpr x C57BL/6J)F2 cross, which differs from the cross used here, we have named this QTL Heal25. Table 3 notes this QTL mapping to Chromosome 11 with peak marker D11Mit213 between 58 cM - 70 cM. Increased healing is conferred to male mice inheriting the MRL/MpJ derived allele.

Novel QTLs identified in this study are Heal12 and Heal13. Heal12 maps to 12 cM mouse Chromosome 14 between D14Mit201 (7.5 cM) and D14Mit233 (19.5 cM), LRT=19.8. CAST/EiJ-derived alleles confer dominantly inherited increase in wound healing at Heal12. Heal12 is a male associated locus. Ang1 (18 cM) and Ang2 are suggested as possible candidate genes for Heal12.

Heal13 mapped to 44 cM mouse Chromosome 17 between 35 cM and 45 cM (LRT=19.3, LRS>22). CAST/EiJ-derived alleles confer additively inherited increase in wound healing at Heal13. This locus shows a stronger effect in male animals. Ltbp1 is suggested as a potential candidate gene for Heal13.

Heal12 and Heal13 show strong epistatic interaction.
