Mutation details: A loxP site was inserted upstream of exon3 and an FRT flanked neo cassette and loxP site were inserted downstream of exon 3 via homologous recombination. Flp mediated recombination removed the neo cassette. Cre mediated recombination removed exon 3. Autoradiography revealed loss of functional protein, as reflected by binding of the highly selective OPRK1 agonist [3H]U69,593. Quantitative reverse transcription PCR (qPCR) analysis confirmed loss of mRNA expression throughout the brain of homozygous mutant mice.
