Mapping and Phenotype information for this QTL, its variants and associated markersJ:18261The backcross (NZB/BlNJ x NZW/lacJ)F1 x NZW/lacJ was screened for approximately 350 SSLP markers. There was a significant association with the expression of autoimmune renal disease and distal Chromosome 4 markers. Using x2 analysis of Nba1, there were6 markers which gave a significant P value (in parenthesis): D4Nds2 (0.005) - D4Mit11 (0.005) - Lck (0.001) - D4Mit16 (0.005) - Epb4.1 (0.0005) - D4Mit48 (0.001). The authors place Nba1 7 cM distal to Lck, between Epb4.1 and D4Mit48. This was based on 7 of the backcross mice with a recombinant between Elp1 and D4Mit48. J:36093Linkage analysis was performed on a population of female (NZB/BlNJ x NZW/LacJ)F1 x NZW/BlNJ backcross animals to identify QTLs associated with autoimmune nephritis traits. Neither parental strains NZB/BlNJ nor NZW/LacJ develop severe nephritis. However, (NZB/BlNJ x NZW/LacJ)F1 hybrid animals do develop severe nephritis with several autoantibody traits. This study extends a previous nephritis mapping experiment using 60 female animals from the same kind of cross. In this study 48 additional female animalsand 9 microsatellite markers were added to the analysis. Suggestive linkage to nephritis mapped to mouse Chromosome 4 between Epb4.1 (65.7 cM, P=0.001) and D4Mit48 (69.8 cM, P=0.0006). This locus colocalizes with Nba1, a previously mapped autoimmunity QTL. A potential candidate gene for this locus is C1qa (66.1 cM).07.14.2015 Curator Note: Because Nba1 was originally mapped in J:18262 in 1994 using an identical cross to the backcross used here, and it mapped to the same genomic segment, we consider the QTL identified here to be the same QTL, Nba1. |
