Mutation details: A cDNA clone was produced using RNA purified from a confirmed alveolar soft part sarcoma tumor. The cDNA contained the ASPCR1/TFE3 fusion product of the t(X;17) chromosomal translocation typical of these tumors. This cDNA was inserted downstream of a floxed STOP sequence consisting of the neomycin resistance gene and a polyadenylation transcription termination sequence and upstream of an IRES-EGFP. Both the fusion gene and EGFP will be translated form the same transcript following cre-mediated recombination to remove the STOP.
