Mutation details: An S1700A mutation followed by an FRT flanked neo cassette were inserted in exon 41 via homologous recombination. Flp mediated recombination removed the neo cassette. The Ala substitution for Ser-1700 does not substantially affect native channel expression or distribution, the stoichiometry of the distal (dCT) and proximal C terminus (pCT) in the autoinhibitory complex, or the overall beta-adrenergic signaling, as assessed by phosphorylation of S1928 in response to isoproterenol.
