Mapping and Phenotype information for this QTL, its variants and associated markers

J:73060

Composite interval mapping (CIM) was used for localization of QTL involved in modulating response to infection with Borrelia burgdorferi. Parental strains C3H/HeN and C3H/HeJ are susceptible to B.burgdorferi-induced arthritis whereas parental strains C57BL/6N and BALB/cAnN are resistant. Four different backcrosses and the reanalysis of a previous intercross were performed.

In the (C57BL/6N x C3H/HeN) x C3H/HeN backcross a QTL is identified on Chr 17. Authors equate the this QTL with a previously identified QTL Bbaa8.

10.28.2105 Curator Note: Since Bbaa8 was originally identified in J:52017 using an (C3H/HeNCr x C57BL/6NCr)F2 intercross which differs from the population of the backcross used here we condiser the current study a separate mapping experiment and have named

the QTL Bbaa28.

Bbaa28 was linked to both specific IgG (LRT=40.2) and IL6 (LRT=35.4) from 10-23.3 cM mapping between markers D17Mit46 and D17Mit215 accounting for 14.0% and 14.3% of trait variance, respectively.

In the (C3H/HeJ x BALB/cAnN) x BALB/cAnN backcross another QTL was identified on Chr 17.

This QTL was also equated with Bbaa8 and was renamed by curators to Bbaa29 because of the differing mapping populations. Bbaa29 was linked to ankle swelling at 22.5 cM at marker D17Mit176, LRT=14.5 accounting for 11.3% of trait variance. It was also linked to specific IgG at 24.cM with marker D17Mit10, LRT=12.8 accounting for 9.7% of trait variance. [Table 2.]

The reanalysis of the (C57BL/6N X C3H/HeN)F2 intercross confirmed the previoulsy identified Bbaa8 QTL on Chr 17. Using CIM Bbaa8, linked to total IgG, mapped from 22.5 - 23.3 cM between markers D17Mit176 and D17Mit215, LRT=19.4. [Table 3.]

C3H/HeN-derived alleles are associated with increased arthritis susceptibility and severity at all QTL mapped in this experiment.
