Mutation details: A concatemer of sleeping beauty transposable elements each consisting of a floxed PGK-neo cassette followed by inverted repeat direct repeat sequence specific for sleeping beauty transposase flanking human BCL2 splice acceptor sequence, IRES, lacZ, poly A signal, a chimeric CAG promoter containing sequence from the chicken ACTB promoter, GFP, and splice donor sequence. Expression of GFP is not seen in the concatemer. Transposition inside a gene should result in ubiquitous expression of GFP by the CAG promoter and expression of lacZ driven by the endogenous promoter. Transposition by sleeping beauty transposase does not move the PGK-neo cassette. The concatemer inserts into chromosome 10.
