Two mapping experiments were performed to identify the genetic loci that regulate IGF-1.

IGF-1 levels vary significantly between two inbred strains with MRL/MpJ mice having the much higher plasma IGF-1 level then SM/J mice. In the first analysis an (MRL/MpJ x SM/J) F2 intercross was performed. 136 female and 235 male mice were used. Using sex as an additive co-variate three main effect QTL were identified on Chr 10, 9 and 17.

A significant QTL, Igf1q4, mapped to Chr 10 peaking at 43 cM nearest marker rs6394370 with a LOD=31.8. The 95% confidence interval ranged from 42-44 cM. The MRL/MpJ allele conferred higher IGF-1 levels in an additive mode of inheritance. [Table 2.]

A suggestive QTL, Igf1q5, mapped to Chr 17 peaking at 10 cM nearest marker rs6384940 with a LOD=5.8. The 95% confidence interval ranged from 8-58 cM. The MRL/MpJ allele also conferred higher IGF-1 levels in an additive mode of inheritance at this locus. [Table 2.]

Another suggestive QTL, Igf1q3, mapped to Chr 9 peaking at 26 cM nearest marker rs13480173 with a LOD=4.1. The 95% confidence interval spanned between 18-36 cM. The MRL/MpJ allele conferred higher IGF-1 levels when inherited recessively at this locus. [Table 2.]

One female-specific QTL, Igf1q6, mapped to Chr 15 peaking at 40 cM nearest marker rs3681229 with a LOD=3.7. The 95% confidence interval ranged between 22-46 cM. The MRL/MpJ allele conferred higher IGF-1 levels when inherited recessively at this locus. [Table 2.]

In the second analysis, haplotype analysis (HAM) among 28 domesticated inbred strains (described in J:216124) identified three suggestive QTL in female mice on Chrs 2, 10 and 17 and four male mice on Chrs 1, 3 and 16.

Female specific QTL:

Igf1q7 mapped to Chr 2 between 153602815 and 153638312 bp on NCBI build 37 with a LOD=3.67;

Igf1q8 mapped to Chr 10 between 88735006 and 89031602 bp on NCBI build 37 with a LOD=4.12 and;

Igf1q9 mappped to Chr 17 between 28830413 and 28853776 bp on NCBI build 37 with a LOD=3.77. [Table 2.]

Male specific QTL:

Igf1q10 mapped to Chr 1 between 158911588 and 159733488 bp also on NCBI build 37 with a LOD=5.14;

Igf1q11 mapped to Chr 3 between 52448663 and 52821781 bp on NCBI build 37 with a LOD=4.05;

Igf1q12 mapped to Chr 3 between 67787738 and 69548776 bp on NCBI build 37 with a LOD=4.44 and;

Igf1q13 mappped to Chr 16 between 74123988 and 74157921 bp also on NCBI build 37 with a LOD=3.84. [Table 2.]
