Mutation details: Exon 33 was replaced with a floxed neomycin resistance cassette and a modified exon in which a nucleotide substitution (A to G) results in the amino acid substitution of arginine for lysine at position 1646 (K1646R). Cre-mediated recombination removed the selection cassette. Kinase assays showed that native kinase activity was abolished in mouse embryonic fibroblasts and peritoneal macrophages derived from homozygous mutant mice, whereas protein levels remained unchanged relative to wild-type controls.
