Mapping and Phenotype information for this QTL, its variants and associated markersJ:221321This study proposes to remap known QTL Tir1 (Chr17) and Tir2 (Chr 5) and Tir3 (Chr1), originally mapped using (BALB/cOlaHsd x C57BL/6JOlaHsd)F2 and (A/JOlaHsd x C57BL/6JOlaHsd)F1 intercrosses (J:40771), and combining additional mapping with haplotype analysis, aCGH, and resequencing to reduce a long list of candidate genes within these QTL regions to a short list of candidate genes with defined genetic differences that correlate with phenotype. An additional cross of 345 (C57BL/6JOlaHsd x C3H/HeJOLsHsd) F2 mice were phenotyped for survival time after infection with T. congolense, strain IL1180. Ninty four of the animals with extreme survival times were selectively genotyped with mircostaellite and SNP markers across the three known QTL, Tir1, Tir2 and Tir3. Mean survival times post infection for the parental founder strains were 63 days for the C3H/HeJ and 87 days for the C57BL/6J.05.10.2016 Curator Note: Because Tir1, Tir2 and Tir3 were orginally mapped using a (BALB/cOlaHsd x C57BL/6JOlaHsd)F2 intercross, a mapping population that differs from the one used here, we have named QTL identified here using the (C57BL/6JOlaHsd xC3H/HeJOLsHsd) intercross as Tir7 (Chr17) and Tir8 (Chr1).QTL Tir7, trypanosome infection response 7, mapped to Chr 17 with peak marker D17Mit81 at 16 cM with a LOD=6.344 in a 95% confidence interval that spanned 17 cM. [Table 2].QTL Tir8, trypanosome infection response 8, mapped to Chr 1 with peak marker D1Mit356 at 94.9 cM with a LOD=3.614 in a 95% confidence interval that spanned 47 cM. [Table 2].C3H/HeJ alleles reduce survival time at both Tir7 and Tir8. No QTL was detected on Chr 5 in the region previously mapped identifiying QTL Tir2 in the (BALB/cOlaHsd x C57BL/6JOlaHsd)F2 intercross. |
