Mutation details: The L1L2_Pgk_P cassette was inserted at position 89389112 of Chromosome 15 upstream of the critical exon 9 (Build GRCm39). The cassette is composed of an FRT site followed by lacZ sequence and a loxP site. This first loxP site is followed by neomycin resistance gene under the control of the PGK promoter, SV40 polyA, a second FRT site and a second loxP site. A third loxP site is inserted downstream of the targeted exon 9 at position 89389949. The critical exon 9 is thus flanked by loxP sites. A "conditional ready" (floxed) allele was created by flp recombinase expression in mice carrying this allele. Subsequent cre expression resulted in a knockout mouse. Western blot analysis of brain lysates confirmed the complete absence of isoform a (longest variant) but detected expression of isoforms c/d and e.
