Anxiety and depression are heterogeneous and complex diseases with a high rate of comorbidity. The authors tested the behavioral sensitivity of 543 (C57BL/6J x C3H/HeJ) F2 intercross mice in paradigms for anxiety and depression. All animals were genotyped with 269 microsatellite markers with a mean distance of 5.56 cM.

Trait anxiety was assessed by the zero maze and the lightdark test. In both tests, time spent (s) and distance travelled (cm) in open, lighted areas were measured. State anxiety levels were detected by the startle response test and the animals startle response (g) before as well as after an acoustic stimulus was recorded.Depression-like behavior was determined with the forced swim test by measuring the time (s) the animal was immobile showing no flight behavior. Each animal was analyzed once in all behavioral tests and was left undisturbed for 7 days between two experiments. The behavioral examination was achieved in the following order: (i) zero maze, (ii) lightdark test, (iii) startle response test, (iv) forced swim test.

QTL analysis revealed several new QTL on Chromosome 5 with a common core interval of 19 Mb. The authors further narrowed this interval by comparative genomics to a region of 15 Mb. A database search and gene prioritization revealed the protein coding gene Enoph1 as the most significant candidate gene on the prioritization list for anxiety and also for depression fulfilling the authors' selection criteria. The Enoph1 gene, which is involved in polyamine biosynthesis, is differently expressed in the parental strains C57BL/6J and C3H/HeJ, which have different brain spermidine levels and show distinct anxiety and depression-related phenotypes.

QTL Axtzm1 (anxiety-zero maze 1, time) maps to Chromosome 15 from 8.7 - 25.7 cM with a peak LOD score of 4.59 near 22.2 cM.

QTL Axtzm2 (anxiety-zero maze 2, distance) maps to Chromosome 5 from 49 - 65 cM with a peak LOD score of 4.84 near 61 cM.

QTL Dfsit1 (depression-forced swim 1, immobility time) maps to Chromosome 5 from 41 - 58 cM with a peak LOD score of 14.41 near 53 cM.

QTL Axtldb33 (anxiety-light dark box 33, time) is significant for males only. It maps to Chromosome 5 from 44 - 62 cM with a peak LOD score of 3.69 near 57 cM.

QTL Axtldb34 (anxiety-light dark box 34, time) is significant for males only. It maps to Chromosome 14 from 38.5 - 58.5 cM with a peak LOD score of 4.42 near 55.5 cM.

QTL Axtldb35 (anxiety-light dark box 35, distance) is significant for males only. It maps to Chromosome 1 from 50 - 85 cM with a peak LOD score of 4.79 near 58 cM.

QTL Dfsit2 (depression-forced swim 2, immobility time) is significant for males only. It maps to Chromosome 5 from 42 - 59 cM with a peak LOD score of 6.53 near 52 cM.

QTL Dfsit3 (depression-forced swim 3, immobility time) is significant for males only. It maps to Chromosome 7 from 1.5 - 8.5 cM with a peak LOD score of 4.02 near 3.4 cM.

QTL Dfsit4 (depression-forced swim 4, immobility time) is significant for females only. It maps to Chromosome 5 from 36 - 60 cM with a peak LOD score of 9.1 near 40 cM.
