Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 88 animals from a SWR x (SWR x NZB)F1 backcross population to identify QTLs associated with lupus phenotypes. (SWR x NZB)F1 hybrid animals are susceptible to lupus nephritis. Animals were sero-typed at 9-12 months of age. 116 autosomal microsatellite markers at an average spacing of 10.6 cM were used for the genome scan.

Suggestive linkage to IgM or IgG anti-nuclear antibody (ANA) production mapped to 25 cM on mouse Chromosome 10 near D10Mit15 (LOD=2.11). This locus is tentatively named Swrl4 and also shows weak linkage to glomerulonephritis (LOD=1.1). SWR-derived alleles at Swrl4 confer increased IgM or IgG ANA production with a recessive mode of inheritance.

Suggestive linkage to early mortality mapped to 4 cM on mouse Chromosome 10 near D10Mit146 (LOD=2.17). NZB-derived alleles at D10Mit146 confer susceptibility to early mortality with a dominant mode of inheritance. A locus on distal mouse Chromosome 10 showed weak linkage to IgG3 antibody levels near D10Mit35a at74 cM (LOD=1.76). NZB-derived alleles at D10Mit35a confer increased IgG3 antibodies with a dominant mode of inheritance.
