Mutation details: The transgene has (from 5' to 3') a CMV-IE enhancer/chicken beta-actin/rabbit beta-globin hybrid promoter (from CAGGS), a loxP-GFP-3xSTOP-loxP cassette and a cDNA sequence encoding human EIF4EBP1 rendered constitutively active and mTORC1-nonresponsive by threonine-to-alanine mutations at two mTOR phosphorylation sites [T37A and T46A]). In the absence of Cre recombinase, mice have widespread expression of GFP, and no human EIF4EBP1 expression.
