Mutation details: This knock-in line at the Gt(ROSA26)Sor locus carries the following genetic elements (listed in order from 5' to 3'): CAG promoter, loxP-3x polyadenylation site-loxP ("lox-stop-lox"), membrane-anchored (via an N-terminal myristoylation site) modified tdTomato with a C-terminal triple-myc tag, 2A cleavage site, histone H2B-GFP fusion with C-terminal triple hemagglutinin tag, bovine growth hormone gene 3'UTR, and Frt-PGK-Neo-Frt. In the absence of Cre-mediated recombination, neither the tdTomato nor the GFP reporter is expressed. Upon crossing to a Cre recombinase-expressing transgene or knock-in, the lox-stop-lox cassette is excised in cells that express Cre, leading to production of membrane-associated tdTomato and nuclear H2B-GFP. The fluorescent proteins can be visualized based on their intrinsic fluorescence, or by immunostaining for the fluorescent proteins and/or their associated epitope tags.
