Mutation details: This allele is generated using somatic cell nuclear transfer (SCNT). Individual (C57BL/6 x 129)F1 donor nuclei from iNKT (invariant natural killer T) cells were isolated and used to replace the mitotic spindle from mouse oocytes. Cells are developed to the blastocyst stage and used to generate embryonic stem (ES) cells. ES cells were injected into recipient blastocysts, and blastocysts implanted into pseudopregnant females. The Tcrb allele selected for propagation from the transferred nuclei had a productive, in-frame recombination of the Vbeta7 gene segments with a unique CDR3 C sequence (CASSLPGHNERLFF).
