Mutation details: A vector containing the following elements was targeted to the host locus: the CAG enhancer/promoter, a loxP site flanked STOP cassette, three copies of the sequence coding for the FLAG tag, the cDNA for the coding sequence of the human TMEM14B gene, an IRES, the EGFP fluorescent reporter gene, WPRE (Woodchuck Hepatitis Virus Posttranscriptional Regulatory Element) sequence and poly(A) sequence. Using Crispr/CAS9 technology, the vector plus Cas9, sgRNAs and template DNA were injected into C57BL/6 zygotes. This creates a conditional-ready allele that will express the human gene and the reporter gene only after cre-mediated excision of the STOP cassette. The human gene does not have a mouse ortholog, as it is a family member unique to primates.
