Mutation details: The targeting construct includes a loxP site inserted into intron 1 and the following inserted into intron 2: loxP2 - inverted FRT1 - loxP1 - FRT2 - inverted neomycin casstte - inverted FRT1 - inverted Syt9 coding sequence - inverted loxP1 - inverted loxP2 - FRT2. The Syt9 cDNA incorporated in the allele (intended to function as a conditional knock-in) is non-functional. Flp-mediated recombination excised the region between the two FRT2 sites, leaving a loxP1 site in intron 1 and the following in intron 2: loxP5171-inverted FRT1-loxP1-FRT2.
