Mutation detailsThe targeting vector was designed to insert a multicistronic reporter into the ATG start codon of the F-box protein 2 (Fbxo2) gene. Specifically, this vector includes (from 5' to 3') an H2B-Venus fusion protein (coding sequence for the human HIST1H2BB gene [histone 1 H2bb] with a C-terminal yellow fluorescent protein sequence), a hygromycin resistance gene flanked with viral P2A oligopeptides, a cre/ERT2 fusion gene (Cre recombinase fused to a human estrogen receptor ligand binding domain), a polyadenylation signal, and a loxP-flanked neomycin resistance (neo) cassette. The construct was electroporated into 129P2/OlaHsd-derived E14tg2a embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts and resulting chimeric mice were bred with Tg(EIIa-cre)C5379Lmgd mice on an FVB genetic background to remove the neo cassette. (J:266597)
