Mutation detailsA loxP site and a neomycin resistance gene cassette (in the same transcriptional direction as the targeted gene) were inserted into intron 1. A second loxP site was inserted into intron 1 of upstream neighbor Peg3 (with which it shares a bidirectional promoter) upstream of the Peg3-DMR (differentially methylated region). RT-PCR experiments showed that the insertion of the neo cassette, which contains a poly(A) signal, dramatically reduces the expression of the targeted gene. (J:249082)
