Mutation detailsA hemagglutinin (HA)-tagged rat cDNA for the cytoplasmic variant of Camk2d is under the control of the 5.5-kb murine alpha-MHC promoter. Immunocytochemical staining confirmed that the expressed transgene is expressed only in the heart and is confined in the cytoplasm of cardiomyocytes. Three lines (TGL, TGM, and TGH) were generated. This TGL line carries 3 copies of the transgene and is the low copy number line. Protein levels are increased 12-fold over wild-type levels but the amount of activated Camk2d is only increased 1.7-fold in hearts. (J:130644)
