Mutation details: Cas9 mRNA and single-guide RNA (sgRNA) targeting murine Mettl4 exon 6 was microinjected into the cytoplasm of zygote-stage embryos to induce deletions. Injected embryos were implanted in CD-1 foster mice and pups screened for deletions. From a single round of microinjection, five mice were born and four contained deletions of exon 6. Three founder mice were selected for mouse line expansion prior to experimental analysis. This allele has deletion of exon 6, which creates a translational frameshift that ablates the catalytic domain, for a predicted methyltransferase knockout (KO) allele.
