Mutation details: The targeting construct PG00252_A_1_B04 inserted upstream of the critical exons 2-3 (Build GRCm39). The cassette is composed of an FRT site followed by lacZ sequence and a loxP site. This first loxP site is followed by a neomycin resistance gene under the control of the human beta-actin promoter, SV40 polyA, a second FRT site and a second loxP site. A third loxP site is inserted downstream of the targeted exons 2-3 at position 44154510. The critical exons 2-3 are thus flanked by loxP sites. Flp-mediated recombination deleted the lacZ and neomcyin resistance gene, generating a "conditional ready" exon 2-3 floxed allele. Subsequent cre-mediated recombination deleted exons 2-3 and generated a knock-out allele.
