Mutation details: The targeting consists of a FREX cassette (containing the rox12 and roxP target sites for the Dre recombinase in direct orientation, separated by a 68 bp spacer) inserted in the 5' untranslated region (UTR) of the Pou4f3 locus, 65 bp before the initiator codon ATG; three repeats of the SV40 early region transcription terminator inserted in the 3' UTR, 564 bp downstream of the STOP codon; the Cre recombinase cDNA in reverse orientation and a second FREX cassette in reverse orientation; and the Neomycin positive selection cassette (PGKNeo) flanked by FRT (Flp recombinase target) sites. The targeting vector was introduced to 129-derived R1 embryonic stem cells. The neomycin cassette was excised with a germline FLP recombinase. Dre-mediated recombination generated inversion-excision at this locus results in the expression of Cre from the Pou4f3 promoter.
