Mutation details: Tryptophan codon 338 (TGG) in exon 11 was changed to alanine (GCG) and phenylalanine codon 339 (TTT) was changed to leucine (CTA) (p.W338_F339delinsAL) and a loxP site flanked neomycin resistance gene cassette was inserted into intron 11. The neo cassette was removed through subsequent Cre-mediated recombination. The mutations lock the encoded peptide in the xanthine oxidase (XO) form, so it cannot be converted into the xanthine dehydrogenase (XDH) form. XO activity represents 100% of total activity in liver homogenates.
