Mutation detailsCysteine codon 995 (TGT) in exon 27 was changed to arginine (CGG) (p.C995R) and a loxP site flanked neomycin resistance gene cassette was inserted into intron 28. The neo cassette was removed through subsequent Cre-mediated recombination. The mutation stabilizes the encoded peptide in the xanthine dehydrogenase (XDH) form so that there is a reduced likelihood that it will be converted into the xanthine oxidase (XO) form. XO activity represents <20% of total activity in liver homogenates. (J:281671)
