Mutation details: The targeting vector was designed to insert a loxP site followed by a frt-flanked neomycin resistance (neo) cassette upstream of the endogenous exon 3 of the CD79B antigen (Cd79b) gene. A polyadenylation (pA) site followed by an F3-flanked puromycin resistance (puro) cassette and a second loxP site was inserted downstream of endogenous exon 6. Following the second loxP site, a region containing exons 3-6, with a mutant exon 5, was inserted. Specifically, the mutant exon 5 has a TAT to CAC missense mutation inserted at position 195 of Cd79b. This results in the tyrosine to histidine mutation Y195H, analogous to Y196H in humans. The election cassettes were removed by crossing to to FLPe transgenic mice.
