Mutation detailsA construct containing a loxP site, FRT-flanked pgk-neo cassette, exon 2 containing two amino acid substitutions (C25A, C62A), and second loxP site was inserted in place of the endogenous exon 2 via homologous recombination. Recombinase-mediated recombination removed the neo cassette and exon 2. Immunoblot analysis confirmed the absence of protein expression in homozygous mice. (J:258944)
