Mutation detailsModeling human variant NM_012062.5: c.1207C>T; p.R403C using a conditional by inversion (COIN) allele design. Exons 10, 11, and 12, were duplicated and inserted in an antisense orientation upstream of the endogenous exon 10 in Dnm1l (NM_001276340.2). A Lox66 site was inserted upstream of the antisense exon 10 and a reverse complement Lox71 site was inserted downstream of the endogenous exon 12. A Lox511 site was inserted upstream of the Lox66 site and a reverse complement Lox511 site was inserted downstream of the antisense exon 12. With cre-mediated recombination, the antisense exons are moved into the sense orientation for expression of the variant-containing exon and the endogenous exons are recombined out of the allele.
