Mutation description

• Allele Type: Targeted (Null/knockout)
• Mutation: Intragenic deletion
• Mutation details: A loxP site was inserted upstream of exon 5 and an frt-flanked neomycin selection cassette and second loxP site were inserted downstream of exon 6. The neomycin selection cassette was removed via FLP-mediated recombination in the germline. Further Cre-mediated recombination resulted in the deletion of exons 5 and 6, causing a frameshift and introduction of a premature stop codon. RT-PCR demonstrated absence of mRNA expression in the heart and skeletal muscle while immunoblot analysis confirmed the absence of protein expression in spleen, lung, brain, heart and muscle tissue from homozygous mutant mice.
