Abnormal thymus architecture in Zeb1tm1Yhi/Zeb1tm1Yhi mice

mortality/aging
• about 80% die within 2 days after birth, however the remaining 20% survive to adulthood

immune system
• poorly developed thymus with no distinction of cortex and medulla
• total cell number of thymus is reduced from 1/100 to 1/500 of the heterozygous littermate
• the early stage intrathymic c-kit+ T precursor cells are largely depleted
• development from double negative (DN) to double positive (DP) cells appears to be partially inhibited
• in peripheral lymph node tissue the CD4-CD8+ single positive cells are significantly reduced relative to CD4+CD8- single positive cells
• the relative proportion of CD4+CD8+ cells is reduced from 88% in controls to 57% in homozygous thymocytes while that of single positive cells was increased from 11 to 38%
• the number of lymphocytes recovered from a pair of inguinal lymph nodes is reduced to about 1/10 of that from heterozygous littermates
• reduction of peripheral T cells in the spleen and lymph node
• number of thymocytes at E18.5 is reduced 10-fold
• the relative proportion of CD4+CD8+ cells is reduced from 88% in controls to 57% in homozygous thymocytes
• in peripheral lymph node tissue the CD4-CD8+ single positive cells are significantly reduced relative to CD4+CD8- single positive cells

vision/eye
• iridocorneal adhesions lead to loss of the iridocorneal angle
• iridocorneal adhesions are observed in 4 month old mice
• corneolenticular and iridolenticular adhesions are also evident
• the cornea of 4 month old mice is more than twice as thick as controls
• there number of keratocytes found in the cornea are doubled in 4 month old mice compared to age-matched controls
