renal/urinary system
• formation of mesenchymally derived epithelial structures in explants is compromised compared to wild-type explants
• formation of polarized epithelia from kidney explants is reduced compared to wild-type explants
• kidney explants exhibit 40% fewer laminin+, mesenchymally derived structures compared to wild-type explants
• well-developed S-shaped structures are located farther from the tips of the ureteric buds and have laminin+ fibers separating the structures unlike in wild-type mice indicating non-continuous tubules
• 73.3% of ureteric buds are not fused to the mesenchymally derived epithelia compared to 21% in wild-type mice
• deposition of a well-defined basement membranes by mesenchyme aggregates in the developing kidney is impaired
• however, cellular proliferation within the developing kidney is normal
|
• adult mice have one third fewer nephrons than in wild-type mice due to postnatal necrotic loss
|
• at P1, 23% of nephrons are necrotic compared of 7% in wild-type nephrons
• necrosis of nephrons occurs shortly after the onset of filtration
• however, there is no apoptotic loss of nephrons
