• mice have few (15-20% of wild-type) mature CD8+ T cells in the periphery, but significantly more than Tap1-null animals
• mice mount poor CTL responses to influenza virus, but normal responses to LCMV and vaccinia viruses
• cells infected with an influenza virus or a vaccinia virus minigene encoding an ovalbumin peptide were poorly recognized by nucleoprotein-specific Db-restricted T cell hybridoma or by cytotoxic T lymphocytes, respectively
• expression is 10-15% of that observed on wild-type cells, or 2 times the level on Tap1-deficient cells
• mice display defective assembly of class I peptide-loading complexes
• in null cells, most heavy chains are thermolabile indicating lack of high-affinity peptides; most MHC class I molecules lack high-affinity peptides but traffic normally to the cell surface
