• homozygotes are found at the expected Mendelian frequency at E6.75 through E8.5; however, no homozygotes are obtained after E10.5

• at E7.5-E8.5, mutant embryos exhibit impaired formation of embryonic mesoderm and endoderm; instead, they largely consist of anterior neuroectoderm resulting in a "head-without-trunk" phenotype
• homozygotes fail to exhibit rotation of the anterior-posterior axis, resulting in mislocalization of the head- and trunk-organizing centers
• at E6.75, mutant embryos show absence of embryonic mesoderm; in contrast, extra-embryonic mesoderm is often present, with an allantois and blood islands of the visceral yolk sac mesoderm detected at later stages (E7.5-E8.5)
• at E7.5-E8.5, homozygotes exhibit distally located ectopic folds of neuroectodermal tissue
• at E6.75, homozygotes show absence of a primitive streak
• at E7.5-E8.5, homozygotes display absence of somite formation
• at E6.75, homozygotes display a thickened layer of visceral endoderm at the distal tip of the egg cylinder

• these changes in expression indicate defects in axial mesendoderm and definitive endoderm development with a nearly complete loss of definitive endoderm in severely affected embryos
