cardiovascular system
• double homozygotes are hypotensive compared to wild-type, however blood pressure is increased compared to Agtr1tm1Afu homozygotes

homeostasis/metabolism
• mice exposed to acid aspiration to induce acute lung injury show improved lung function, reduced edema formation, and greatly attenuated pulmonary vascular permeability compared to wild-type control

renal/urinary system
• reduced interstitial volume

N normal phenotype
cardiovascular system
• homozygotes display a 22 mm Hg reduction in systolic blood pressure (77% of wild-type values) (J:28041)
• after central injection of angiotensin II, adult male homozygotes exhibit a significantly smaller increase in SBP than wild-type males within the range of 50-200 ng, with no differences in the time course of the pressor response; no obvious dose dependence is observed, although a dose of 200 ng results in a slightly increased SBP (J:107851)
• i.c.v. injection of angiotensin II (100 ng) has no effect on SBP in male homozygotes; also, PD-123319 does not significantly change BP after central injection of angiotensin II (J:107851)
• homozygotes exhibit chronic hypotension, with a significant decline in both the systolic and diastolic blood pressure relative to wild-type mice

homeostasis/metabolism
• homozygotes display hyperreninemia, with a 7- to 8-fold elevation in the plasma concentration of active renin relative to wild-type and heterozygous mutant mice
• homozygotes show a significant increase in renin mRNA levels in the kidney and in plasma renin activity relative to wild-type mice

behavior/neurological
• i.c.v injection of angiotensin II (100 ng) increases water intake 5-fold in adult wild-type males; this increase is reduced by ~60% in adult male homozygotes relative to wild-type males
• in wild-type males, the effect of angiotensin II on water intake is significantly inhibited by valsartan and by PD-123319; in male homozygotes this effect is suppressed by ~40% by PD-123319 but not valsartan

behavior/neurological
• i.c.v injection of angiotensin II (100 ng) increases water intake 5-fold in adult wild-type males but has no significant effect on water intake in males homozygous for Agtr1atm1Afu and hemizygous for Agtr2tm1Gsb

cardiovascular system
• adult males homozygous for Agtr1atm1Afu and hemizygous for Agtr2tm1Gsb show no apparent physiological changes (e.g. body weight gain) or histological abnormalities in the heart, artery or brain
• however, these mice display a higher increase in SBP after central injection of a high dose of angiotensin II (200 ng) relative to Agtr1atm1Afu homozygotes
