liver/biliary system
• apoptotic bodies are detectable in livers of both sexes at 6-12 months of age while none are observed in control
|
• mice by 1 month of age and continuing through 12 months of age have liver weights that are greater than 10% of total body weights
• male mice have significantly higher liver weights than female transgenic mice, and both sexes have higher liver weights than non-transgenic controls
• by 17 months of age, 50% of male mice and 23% of female mice have liver weights that are over half the total body weight
|
• frequent mitotic bodies and dysplastic cell nuclei are evident in 100% of males and 25% of females under 6 months of age
• nuclear shapes are highly irregular, forming notched shapes, slipper shapes, and in some cases a dumbbell shape that seemed to suggest incomplete mitosis
• some abnormally large hepatocytes contained large nucleoli, nuclear and cytoplasmic inclusions, glycogen bodies, and clumped cytoplasmic material suggestive of Mallory's hyaline
• tripolar mitotic bodies are also evident

• some abnormally large hepatocytes contained large nucleoli, nuclear and cytoplasmic inclusions, glycogen bodies, and clumped cytoplasmic material suggestive of Mallory's hyaline
|
• 31% of mice develop hepatocellular carcinoma by 17 months of age regardless of sex
• measurable lesions range in size from 0.6 mm to 1.7 cm
|
• nodular foci of poorly differentiated hepatocytes are first observed in the liver of 9 month old males
• by 12 months of age, multifocal eosinophilic and clear cell nodules are apparent in about half the mice of both sexes
• nodules typically ranged in size from 1-2 mm in diameter at 12 months of age
• by 17 months age, 69% of female mice and 87% of male mice have adenomas or hepatocellular carcinoma that developed from adenomas
