immune system

• infection with measles virus (MV) generates a large cytotoxic T cell population that can target human cells presenting MV antigens in the context of HLA-B27
• these HLA-B27-restricted CTLs have high affinity for MV antigens with half maximal CTL-mediated lysis of target cells occurring at low (3x10 M-8) peptide concentrations
• these CTLs fail to generate a cytotoxic response against target cells with HLA-A2 haplotype

immune system
|
• 8-9% of splenocytes express human CD8alpha
• about 80% express human CD8alphaCD8beta heterodimers while about 20% express CD8alpha-alpha homodimers
• either CD4+ or CD8+ T cells can express the human CD8 complex in the periphery
|
|
• expression of human CD8 allows for effector cell function against target cells presenting viral peptide on human HLA-A2 complexes
• this cytolytic effector function is primarily carried out by T cells expressing both mouse and human CD8 complexes
• the presence of the human CD8 co-receptor does not interfere with the ability of T cells to recognize mouse MHC
