• after superovulation many oocytes fail to progress to metaphase II and are either degenerated or arrested at the germinal vesicle stage
• after 3 hours in culture only 50% of oocytes progress from the germinal vesicle stage to germinal vesicle breakdown compared to 79% of heterozygous oocytes
• addition of 100 uM Rp-adenosine 3,5-cyclic monophosphorothioate rescues maturation of homozygous oocytes
• homozygous oocytes are more sensitive to inhibition of germinal vesicle breakdown by phosphodiesterase inhibitors or cAMP analogs
• in crosses to wild-type males homozygous females have a birth rate of 0.26 litters/female/month and a litter size of 2.2 +/- 0.8 pups/litter compared to 0.86 litters/female/month and a litter size of 7.7 +/- 1.2 pups/litter for heterozygous females
• in crosses to wild-type males homozygous females have a litter size of 2.2 +/- 0.8 pups/litter compared 7.7 +/- 1.2 pups/litter for heterozygous females
