• Background Sensitivity: unlike mice on a mixed C57BL/6J and 129S1/Sv background which display normal body weight throughout life mice on an inbred C57BL/6J background display a decrease in body weight that becomes more pronounced with age
• significant decrease at 6 months of age but not at 3-5 months of age
• however, no difference is seen in body length

adipose tissue
• reduction in body weight is primarily the result of reduced fat mass

homeostasis/metabolism
• in males and females at 10 months of age
• in males and females at 10 months of age
• 3-fold decrease at 10 months of age
• elevated core body temperature in 12 month old mice

• no significant differences in 24 h food intake under normal conditions
• following a 24 h fast mice do not display increased food intake at 1 or 4 hours post fat but do show a slight increase in intake at 24 h post fast, unlike wild-type mice which display small but significant increases in food intake at 1, 4, and 24 h post fast
• intraperitoneal injection of 3 mg/kg melanotan II inhibits food intake for a longer time compared to wild-type mice
• repeated administration of 3 mg/kg melanotan II over 4 days prevents weight gain during this period in mutant but not wild-type mice
