vision/eye
• after 48 hours exposure to 3000 lux light, nuclear layer around central area is reduced
• rod outer segments (ROS) around central area of retina are reduced in length compared to controls
• ROS are shortened in mutants after exposure to 3000 lux light for 48 hours, suggesting loss of photoreceptor outer segments
• delay in dark adaptation indicates clearance of all-trans-retinal is slower than production of 11-cis-retinal in mutants
• after single light flash (~35% bleaching), reduction kinetics of all-trans-retinal are slower than in wild-type; 11-cis-retinal formation is accelerated
• chromophore recovery after intense bleach (~90% of total rhodopsin), 11-cis-retinal production recovery begins after 2 hours compared to 3 hours in wild-type
• after 48 hours exposure to 3000 lux light, total retinoids in mutant eyes are lower than in wild-type
• A2E fluorescent product (from condensation of all-trans-retinal) is detected at much higher levels than in wild-type but lower levels than Abca4tm1Ght mice
• strong light (10000 lux) induces apoptosis (light damage, LD) in mutants after 20 minutes whereas 2 hours is required to induce LD in 129/SvEv controls; only a few cells in C57BL/6 are apoptotic after 2 hours
• extensive LD over a large area is evident with DNA fragmentation after 2 hours of light exposure in mutants
• recovery of a-wave response after bleaching is slower compared to wild-type
• recovery after bleaching is slower than in wild-type
