• mutant males display normal testis morphology and sperm production
• during in vitro fertilization, sperm released from mutant cauda epididymis show an increase in abnormal coiled and bent tails with hairpin structures
• although the size of lumen is reduced it is still filled with strongly eosinophilic secretions, suggestive of partly preserved epithelial secretory function
• in the proximal region, the epithelial cells of mutant seminal vesicles appear to be quite low, cuboidal, and with very little cytoplasm, while the epithelial layer is less folded than normal
• in the distal region, the epithelium exhibits a normal, tall secretory morphology, with occasional small foci of hyperplastic epithelial cells and a decrease in the size of the acini
• the stromal smooth muscle layer appears disorganized and thinner than normal
• at 8 weeks of age, the size of mutant seminal vesicles is only 55% of that in control littemates
• at 8 weeks of age, the weight of seminal vesicle secretion and emptied gland is significantly reduced
• despite normal testis sperm production, sperm numbers are reduced in the caput, normal in the corpus, but significantly increased in the cauda epididymis, suggesting altered sperm epididymal transit kinetics
• mutant epididymi display stromal thickening in the cauda region
• mutant epididymi display stromal thickening in the corpus region
• at 4 hrs after coitus, plugs are detected in equivalent proportions of females mating with control or mutant males; however, plugs formed by mutant males are significantly smaller, lighter, and softer with a fibrous consistency
• the few mutant males that sired a litter yielded an average of 6.2 1.8 pups/litter relative to 9.6 2.7 pups/litter for control males
• unlike control males which yielded an average of 3.7 0.4 litters over a 90-day mating period, only 5 of 15 mutant males (33%) had a single litter, and only 1 of 15 had two litters within the same mating period
• median time to the first litter was 21 days and 42 days for second litter for control males, compared with >90 days for both first and second litters for mutant males
• mutant cauda epididymal sperm show an increased rate of spontaneous acrosome reaction relative to wild-type sperm; hyperactivated, more erratic, and whiplash-like movement typical of capacitated sperm are observed
• in vivo fertility, as measured by oocytes fertilized by mutant males mated with hyperstimulated females at 6 hrs after copulation, is significantly reduced
• in vitro fertilization capacity of mutant cauda epididymal sperm is significantly reduced at lower sperm concentrations (<50,000 sperm/ml) but not at higher sperm concentrations (>50,000 sperm/ml), indicating a mild defect in sperm binding to the zona pellucida that can be overcome by higher sperm numbers

behavior/neurological
• after overnight mating with mutant males, the proportion of plug positive females is significantly reduced, probably due to production of malformed and smaller plugs that are more readily dissolved or dropped
• however, at 4 hrs after coitus, plugs are detected in equivalent proportions of females mated with control or mutant males, indicating successful mating

endocrine/exocrine glands
• although the size of lumen is reduced it is still filled with strongly eosinophilic secretions, suggestive of partly preserved epithelial secretory function
• in the proximal region, the epithelial cells of mutant seminal vesicles appear to be quite low, cuboidal, and with very little cytoplasm, while the epithelial layer is less folded than normal
