• the majority of double-positive (DP) thymocytes fail to upregulate CD5 and CD2, which are indicators of pre-TCR signaling
• cultured DP thymocytes upon stimulation fail to lower expression of CD4 and CD8, and increase expression of Nur77
• there is a partial defect in transitioning from DN3 cells to DN4 cells
• there is a significant skewing towards CD4 single positive T cells in the ratio of CD4:CD8 T cells in the thymus
• the frequency and absolute number of NKT in the thymus is drastically reduced
• the number of NKT cells is also reduced in the spleen and liver
• expression of NK1.1 on the remaining NKT cells in thymus, liver, and spleen is markedly diminished
• DP and CD4 single positive thymocytes flux less calcium upon in vitro activation
