immune system
• bone marrow derived dendritic cells (BMDC) have an augmented ability to present antigen in vitro to both CD4+ and CD8+ T cells
• co-culturing of OVA-specific wild-type T cells and mutant BMDCs leads to increased proliferation and IL-2 secretion by the T cells
• similar results occur with BMDC stimulation of allogenic T cells
• these enhanced responses are dependent on direct cell-to-cell contact
|
• mutant mice that are sub-lethally irradiated and then receive allogenic splenocytes all die within 10 days compared to wild-type recipients where half the group live past 20 days after allogenic splenocyte infusion
• T cells isolated from these mice produce 2-3 fold more IFN-gamma when stimulated by allogenic cells in vitro compared to controls
