Aberrant mitoses in Cdc20tm1.1Pzg/Cdc20tm1.1Pzg MEFs

mortality/aging

cellular
• in cultured MEFs as early as passage 3 the percentage of cells with abnormal karyotypes in increased compared to wild-type cells
• in MEFs, in metaphase chromosomes are misaligned and in anaphase lagging chromosomes or chromosome bridges are seen
• anaphase begins prematurely
• cell growth in cultured MEFs stops with increasing passage numbers as the result of massive cell death
• cell death can be rescued by immortalizing MEFs with the expression of the SV40 large T antigen
• very high levels of apoptotic cell death are seen in embryos
• MEFs grow more slowly than controls

embryo
• very high levels of apoptotic cell death are seen in embryos

Liver tumor of a Cdc20tm1.1Pzg/Cdc20+ mouse

cellular
• in cultured MEFs as early as passage 3 and in splenocytes from adult mice the percentage of cells with abnormal karyotypes in increased compared to wild-type controls

neoplasm
• palpable tumors can be detected by 6.8 months of age and by 24 months half of mutants have tumors compared to only 10% of wild-type controls
• of mice with tumors, 38% have tumors at multiple organ sites
• 4 of 29 tumors were hepatomas
• 25 of 29 tumors were lymphomas

liver/biliary system
• 4 of 29 tumors were hepatomas
