Del(15Krt7-Krt18)1Tmm/Del(15Krt7-Krt18)1Tmm embryos exhibit severe growth retardation and apoptosis

mortality/aging

embryo
N
• embryonic and extraembryonic epithelia is intact
• 10-fold increase in apoptotic cells in embryos, indicating a nutrition defect
• starting around E8.5, fully apparent by E9.5
• desmosomes are smaller and partially mislocalized

homeostasis/metabolism
• expression analysis indicates a nutrient shortage exists in embryos at E9.5
• addition of exogenous glucose fails to rescue the apparent nutrient shortage
• at E9.5 protein biosynthesis is reduced in the embryo and in the yolk sac compared to wild-type controls

cellular
• 10-fold increase in apoptotic cells in embryos, indicating a nutrition defect

growth/size/body
• starting around E8.5, fully apparent by E9.5

neoplasm
• mice treated with an adenovirus expressing Cre recombinase (AdCre) develop lung tumors within 6 weeks of AdCre treatment
• keratin-deficient tumors are typical papillary adenomas and adenocarcinomas, exhibit similar proliferation and apoptosis rates and expression of EMT and neuroendocrine markers as seen in tumors from single Tg(Actb-KRAS*G12V,-luc)#Lche mice
• tumors in AdCre treated mice are multiple papillary adenomas and invasive papillary adenocarcinomas
