N normal phenotype
mortality/aging
• starting at 8 weeks, mice die suddenly with only 10% of mice reaching the age of 30 weeks

nervous system
N
• mice exhibit normal brain morphology
• following middle cerebral artery (MCA) occlusion, mice exhibit reduced infarct volume and improved Bederson scores for global neurological function and grip strength compared with similarly treated wild-type mice
• mice transplanted with wild-type bone are still protected from middle cerebral artery (MCA) occlusion neuronal damage
• following middle cerebral artery (MCA) occlusion, mice exhibit a 40% reduction in infarct volume compared with similarly treated wild-type mice
• mice subjected to MCA occlusion do not exhibit an increase in infarct volume over time unlike similarly treated wild-type mice
• capacitive calcium ion entry (CCE) is reduced compared to in wild-type neurons
• neurons exhibit lower basal calcium ion levels compared with wild-type mice
• peak amplitude of calcium ion release is decreased compared to in wild-type mice
• endoplasmic reticulum calcium ion content and cytosolic calcium ion concentration are decreased compared to in wild-type mice
• during one hour or oxygen-glucose deprivation (OGD), neurons exhibit only a small increase in intracellular calcium ion concentration compared with similarly treated wild-type neurons
• neuron exposed to OGD or reduced pH and/or reduced glucose exhibit increased viability compared with similarly treated wild-type neurons
• however, subsequent calcium ions re-entry after re-addition of extracellular calcium ion is normal and intracellular calcium ion concentration two hours after OGD are markedly increased
• staurosporin-treated neurons exhibit reduced apoptosis compared with similarly treated wild-type neurons
• fewer neurons undergo apoptosis under ischemic conditions compared to similarly treated wild-type neurons
• however, TPEN-treated neurons exhibit normal rates of apoptosis

behavior/neurological
N
• mice exhibit normal swimming ability
• in a Morris water maze, mice take longer to find the hidden platform and exhibit increased total distance in acquisition trials compared with wild-type mice
• following middle cerebral artery (MCA) occlusion

homeostasis/metabolism
• following middle cerebral artery (MCA) occlusion, mice exhibit reduced infarct volume and improved Bederson scores for global neurological function and grip strength compared with similarly treated wild-type mice
• mice transplanted with wild-type bone are still protected from middle cerebral artery (MCA) occlusion neuronal damage
• following middle cerebral artery (MCA) occlusion, mice exhibit a 40% reduction in infarct volume compared with similarly treated wild-type mice
• mice subjected to MCA occlusion do not exhibit an increase in infarct volume over time unlike similarly treated wild-type mice

cellular
• staurosporin-treated neurons exhibit reduced apoptosis compared with similarly treated wild-type neurons
• fewer neurons undergo apoptosis under ischemic conditions compared to similarly treated wild-type neurons
• however, TPEN-treated neurons exhibit normal rates of apoptosis
