• no live-born homozygous animals were recovered in litters from 10 intercrosses of heterozygous mice
• in culture, after 3 days of tamoxifen treatment, ES cells carrying one floxed allele of Tardbp and one null allele expressing tamoxifen-inducible cre recombinase exhibit a reduction in proliferation (colony size) and increase apoptosis, compared to control ES cells with one functional Tardbp allele, with the differences increased by day 9 after induction; these results indicate that Tardbp is essential for ES cell survival and possibly embryonic survival of homozygous embryos
