nervous system
• mice develop spontaneous recurring limbic seizures about 1 week after electrical kindling, although in some cases seizures began before mice were fully kindled
• electrical kindling induces spontaneous recurring seizures after a short latency in about 80% of mice; seizures occur 1-2 times per day and most are bilateral, tonic-clonic seizures
• however, no to little neuronal death is seen
• spontaneous IPSCs show a decrease in amplitude in CA1 neurons and dentate granule cells, with an average decrease of 23% in CA1 neurons
• however, no differences in spontaneous IPSC rise time, decay rate, or frequency are seen

growth/size/body
• males and females exhibit a massive increase in body weight from 4 weeks of age through 7 weeks compared to controls

behavior/neurological
• food intake is greatly increased compared to controls

nervous system
• Pomc1 neurons tend to be hyperpolarized relative to control neurons
• a large increase in amplitude and frequency of spontaneous IPSCs is observed in Pomc1 neurons in slice preparations from mice with conditional deletion of Lepr, indicating that deletion from presynaptic GABAergic neurons increases the inhibitory tone in Pomc1 neurons

homeostasis/metabolism
• animals have significantly elevated fed and fasting blood glucose levels
• animals have significantly elevated fed and fasted serum insulin levels

adipose tissue
• animals display a massive increase in fat mass relative to controls at 10 weeks of age

nervous system
N
• mice show no significant differences in the paired pulse ratio (PPR) recorded from parvalbumin-positive inhibitory interneurons (PV-INs), suggesting normal presynaptic release probabilities; moreover, the AMPA/NMDA ratio is normal in PV-INs
• mice exhibit a smaller median Psd-95-GFP puncta area per dendritic region in PV-INs than control mice
• however, the density of Psd-95-GFP puncta (a proxy of glutamatergic synapse density) is normal in PV-INs
• analysis of visually evoked gamma oscillations in the primary visual cortex (V1) showed that, when mice are presented with a gray screen, the gray/black screen ratio of both mean and peak 55-65 Hz activity is significantly higher than that in control mice, indicating increased power in the narrowband gamma frequency
• mEPSC frequency is severely reduced in parvalbumin-positive inhibitory interneurons (PV-INs) in the primary visual cortex (V1), as recorded at -70 mV in the presence of TTX and gabazine
• however, mEPSC amplitude, half-maximal rise time and half-maximal decay time in PV-INs are equivalent to those in control mice
• mice exhibit decreased glutamatergic synaptic activity in PV-INs

behavior/neurological
N
• both sexes show no significant changes in short-term spatial memory and contextual fear conditioning tests relative to control mice
• following injection of glutamate N-methyl-D-aspartate (NMDA) receptor antagonist MK-801, mice show a significantly greater increase in locomotor activity than similarly treated controls, as determined by the total number of infrared beam breaks in an open field arena during the 30-60 min period post-injection
• both sexes exhibit significantly more rearing events during a 30-min period of open field exploration, indicating an increased exploratory drive
• both sexes make significantly more beam breaks in the center region of an open field arena during the first 5 min of exploration, suggesting reduced anxiety (J:337062)
• in the elevated plus maze (EPM) test, mice exhibit a significantly higher ratio of entries to the extreme end of the open arm vs. entries into the closed arm and spend significantly less time freezing during the 5-min test (J:337062)
• in the hole-board test, male -- but not female -- mice make significantly more nose pokes than controls during a 3-min session (J:337062)
• in the marble burying task, male -- but not female -- mice bury significantly fewer marbles than controls (J:337062)
• both sexes spend significantly less time freezing on the elevated plus maze apparatus during the 5 min test
• both sexes exhibit significantly more rearing events during a 30-min period of open field exploration, indicating an increased exploratory drive
• however, locomotor activity is normal, as determined by the total number of infrared beam breaks in an open field arena during a 90-min period in the dark or a 30-min period in the light

• addition of leptin fails to reduce the frequency of sIPSCs (spontaneous inhibitory postsynaptic currents) in Pomc1 neurons, while in control slices, the frequency is reduced by about 40%
