skeleton
• in culture, primary osteoblasts isolated from calvarial bones of 2-day-old mice show a slight but significant increase in mineralized nodule formation
• however, alkaline phosphatase activity in calvarial osteoblasts is normal
• osteoclast surface / bone surface (Oc.S/BS%) is significantly decreased at 8 weeks, but not at 52 weeks, of age
• in culture, M-CSF and RANKL-induced osteoclast formation is normal in terms of total osteoclast number and large osteoclast formation; also, no change in osteoclast survival is seen at any timepoint after RANKL withdrawal
• medullary cavity diameter is significantly reduced at the femoral diaphysis at 52 weeks, but not at 8 weeks, of age
• however, other femoral cortical bone parameters (cortical diameter, bone volume, and cortical thickness) are normal at both ages
• trabecular bone pattern factor is decreased at the distal femoral and proximal tibial metaphysis at 8 and at 52 weeks of age
• however, no change in trabecular bone pattern factor is noted in the 6th lumbar vertebra at 8 or at 52 weeks of age; trabecular bone thickness is normal at the femoral and tibial metaphysis and in the 6th lumbar vertebra at both ages
• trabecular bone volume/tissue volume (BV/TV%) is significantly increased at the distal femoral and proximal tibial metaphysis at 8 weeks of age, with an even greater increase noted at 52 weeks of age
• however, trabecular BV/TV% is normal in the 6th lumbar vertebra at both ages
• trabecular number is significantly increased at the distal femoral and proximal tibial metaphysis at 8 weeks of age, with an even greater increase noted at 52 weeks of age
• however, trabecular number is normal in the 6th lumbar vertebra at both ages
• trabecular separation is decreased at the distal femoral and proximal tibial metaphysis at 8 and at 52 weeks of age
• however, trabecular separation is normal in the 6th lumbar vertebra at both ages
• trabecular bone mass is markedly increased in older mice
• single labeled perimeter (SL.Pm/B.Pm (%)) is significantly increased at 52 weeks of age
• however, serum levels of PINP (a biochemical marker of bone formation) are normal at 8 and at 52 weeks, and mineral apposition rate (MAR) is normal at both ages
• femur medullary cavity diameter is significantly smaller at 52 weeks of age, suggesting a lower rate of endosteal bone resorption
• however, serum levels of CTX-I (a biochemical marker of bone resorption) are normal at 8 and at 52 weeks

hematopoietic system
• osteoclast surface / bone surface (Oc.S/BS%) is significantly decreased at 8 weeks, but not at 52 weeks, of age
• in culture, M-CSF and RANKL-induced osteoclast formation is normal in terms of total osteoclast number and large osteoclast formation; also, no change in osteoclast survival is seen at any timepoint after RANKL withdrawal

immune system

cellular
• in culture, primary osteoblasts isolated from calvarial bones of 2-day-old mice show a slight but significant increase in mineralized nodule formation

limbs/digits/tail
• medullary cavity diameter is significantly reduced at the femoral diaphysis at 52 weeks, but not at 8 weeks, of age
• however, other femoral cortical bone parameters (cortical diameter, bone volume, and cortical thickness) are normal at both ages

growth/size/body
N
• female mice show normal general appearance and body weight at 8 and at 52 weeks of age
